Estimation of cystine plus cysteine in proteins by the disulfide interchange reaction.

چکیده
The interchange reaction between disulfides in concentrated acid solutions was demonstrated by Sanger (I) in 1953 with model compounds. Ryle and Sanger (2) later studied the formation of mono-dinitrophenyl cystine peptides in the reaction between insulin and di-DNP-cystinel in order to estimate the rate and extent of the interchange reaction. The disulfide interchange reaction was studied from the standpoint of mechanism by Benesch and Benesch (3). They concluded that the mechanism could be represented by the following scheme:

Mechanism of action of pyruvate kinase. Role of sulfhydryl groups in catalytic activity as determined by disulfide interchange.
Evidence is presented that four sulfhydryl residues play a critical role in the activity of pyruvate kinase. Modification of the four most reactive sulfhydryl groups by 5,5’-dithiobis(Z-nitrobenzoate) leads to a disulfide interchange reaction in which a chemically modified but catalytically active enzyme is produced. This form, which contains 5-thio-Z-nitrobenzoate in disulfide linkage to cyste...

To ascertain the upper limit of protein thermostability and to evaluate the introduction of additional -S-S- bridges for the enhancement of protein thermostability by site-directed mutagenesis, the stability of cystine residues at 100 degrees C has been investigated in a number of unrelated proteins. All proteins studied (more than a dozen) undergo heat-induced beta-elimination of cystine resid...
متن کاملThe Photometric Determination of Cystine, Cysteine, Ascorbic Acid, and Related Com- Pounds with Phosphotungstic
Some time ago a brief report (1) was made on the use of the Pulfrich photometer for the determination of sulfhydryl and disulfide compounds by a modified Folin (2) method. Since then the method has been extensively used in this laboratory (3-9). Folin’s method is based upon the observation that cystine plus sulfite gives a blue color with phospho-18-tungstic acid; whereas cystine alone develops...
متن کاملPantethine and cystamine deplete cystine from cystinotic fibroblasts via efflux of cysteamine-cysteine mixed disulfide.
Children suffering from cystinosis, a genetic disease characterized by high levels of lysosomal cystine, are currently being treated with cysteamine to lower the cystine levels in their cells. In fibroblasts from these patients, cysteamine and its disulfide, cystamine, are equally effective in lowering cystine levels. We recently reported that pantethine, a dietary precursor of coenzyme A, depl...

The mechanism of the growth-promoting action of 2-mercaptoethanol on mouse lymphoma L1210 cells in vitro has been studied. The normal line of L1210 is deficient in its capacity to take up cystine, and this limits the growth of the cells in normal culture media containing cystine. In the presence of 2-mercaptoethanol, the cells obtain a high capacity to utilize cystine in the medium. The reactio...
